The facility

Flowcytometry (FCM) is a technique that distinguishes individual particles based on light scatter and fluorescence. A large advantage is that FCM can analyse and sort cells at a very high speed of up to 10,000 cells/second.

In addition to fast cell counts, the individual cell properties are measured. These properties can occur naturally like relative size and pigment composition, or artificially stained like DNA/RNA and enzyme activity. Up to 17 parameters or fluorochrome signals can be measured simultaneously. 

The cells can be sorted based on these parameters, either as single cells or as subpopulations into well plates, tubes or onto Petri dish.

The BD influx flowcytometer at the NIOO is a research machine and flexible in its setup. This makes it an ideal tool to investigate cell populations, typically - but not limited to - phytoplankton, bacterial or viral particles, populations or communities.

Equipment 

The BD INFLUX cell sorter is a flexible and stable stream-in air flowcytometry sorter platform. The instrument has the capability of six-way sorting into tubes or single cell sorting directly into plates or slides and is equipped with five lasers. Furthermore, it is equipped with improved forward scatter (small particle detection down to 200mu) and polarisation sensitive detectors.

  • Lasers: 355nm, 405nm, 488nm, 532nm, and 640nm
  • Nozzle tips: 70µm, 86µm, 100µm, 140µm
  • Controlled temperature environment.

All data are managed according to the NIOO Research data policy, which is in compliance with FAIR data guidelines. Available datasets are available through DRYAD (arising from publications) or upon request (not covered in publications).

Conditions for access

Analysis and sorting on the BD Influx will be supported by an experienced operator (>10 years). Flowcytometry and cell sorting lab is available for collaborative research projects with AqE (NIOO-KNAW), and as such access and costs will be agreed upon by project partners.
